The DNA synthesis of VH and VL sequences

In order to express the variable regions of the heavy chain (VH) and light chain (VL) of an antibody, the VH and VL inserts are subcloned into specific expression plasmids designed for heavy chain (HC) and light chain (LC) production. This process involves inserting the DNA sequences encoding the VH and VL regions into the appropriate expression plasmids, which are specialized circular pieces of DNA that serve as carriers for the genes of interest. The expression plasmids are designed to contain regulatory elements, such as promoters and terminators, which control the expression of the VH and VL sequences in the host cell. This allows for efficient production of the VH and VL proteins.
Expression and Validation of Antibodies at Small Scale in a Mammalian System
Small-scale antibody expression in a mammalian system typically involves introducing the genes encoding the antibody's heavy and light chains into mammalian cells, such as CHO (Chinese Hamster Ovary) cells, using transfection methods.
